Pippin Pulse脉冲场电泳系统

  • 关于脉冲场电泳
        长读长测序(又称三代测序)、合成生物等诸多技术应用都涉及对超长DNA片段的分离与分析。普通的琼脂糖凝胶电泳是借助琼脂糖凝胶的分子筛作用,核酸片段因其分子量或分子形状不同,电泳移动速度有差异而分离。然而这项技术一般只能分离10 kb以下的小分子DNA,更大的DNA分子用普通琼脂糖凝胶电泳无法达到分离的目的,从而限制了染色体DNA的分离、大片段基因组文库构建、生物物种鉴定、病原微生物基因分型、人类染色体内切酶物理图谱分析等方面的研究。而脉冲电场技术通过切换凝胶内的电场方向,能够驱使DNA分子在凝胶内前后方向来回运动,有效地降低大分子核酸片段的迁移速率,同时由于小分子核酸片段改变方向的速率要比大分子核酸片段快,因此通过脉冲场电泳技术,可以有效将大分子DNA片段进行分离和分析。

        美国Sage Science公司的Pippin Pulse脉冲场电泳系统可对长达430kb的超长DNA片段进行高效的电泳分离,而且仪器体积小巧不占空间。用户可通过PC端选择预设程序(如下图)或自定义程序进行大分子DNA片段的脉冲场凝胶电泳分离,程序可自定义使用户的应用更加广泛和灵活。

        使用Pippin Pulse对大分子DNA进行电泳分离后的结果如下图所示:

        2020年发表在Nature Biotechnology上的一篇文献中,研究人员对无角牛进行基因组测序组装和表型分析。其中Pippin Pulse用于牛肝脏基因组提取后的质控以及打断后(50kb)片段分布质控,随后联合Nanopore测序平台进行文库构建和下游测序分析。

        2022年发表在预印本网站BioRxiv Preprint上的另外一篇文献中,研究人员对存在于牛蝇体内的金黄色葡萄球菌引起牛皮肤病的致病机理进行研究。其中Pippin Pulse用于细菌基因组提取后、文库制备前的片段分布质控,随后联合Illumina和Nanopore测序平台进行文库构建和下游测序分析。
